DNA barcoding of Mangroves using Ribosomal ITS Marker in Rhizophoraceae

چکیده
DNA barcode is a very short, standardized DNA sequence in a well-known gene. It provides a way to identify the species to which a plant, animal or fungus it belongs. One of the most popular sequences identification is achieved by comparing DNA sequences of the internal transcribed spacer (ITS) region of nuclear ribosomal cistron. DNA barcoding is a new concept of identifying unknown organisms based on species DNA regions. The nuclear ribosomal Internal Transcribed Spacer region is widely used as a DNA bacoding marker to characterize the diversity and composition of plant groups. The ITS region has been heavily used in both molecular methods and ecological studies, due to its high degree of interspecific variability, conserved primer sites and multiple copy nature in the genome. Primers have long been available for the nuclear ITS (Internal transcribed spacer) rDNA region which are now commonly used for plant identification.

Genomic relations among two non-mangrove and nine mangrove species of Indian Rhizophoraceae.
Random amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP) markers were used to study the genomic relationship among 11 members of Indian Rhizophoraceae represented by nine true mangroves and two non-mangrove species. The AFLP and RAPD bands were scored and analyzed for genetic similarities and cluster analysis was done which separated the 11 species studied into ...
متن کاملIdentification of Medicinal Mangrove Rhizophora apiculata Blume: Morphological, Chemical and DNA Barcoding Methods
"Kandal" (Rhizophora apiculata Blume) is a medicinal mangrove plant used to treat pain, inflammation and reduce blood glucose level in Southeast coast of India. Identification and revision of R. apiculata are largely based on morphological characteristics. Previously, we reported and filed a patent on anti-diabetic Glycosin from R. apiculata. Subsequently, for the quality and market authorizati...
متن کاملPhylogenetic Assessment of Some Species of Crocus Genus Using DNA Barcoding
DNA barcoding is a simple method for the identification of any species using a short genetic sequence from a standard genome section. The present study aimed at examining the nuclear and chloroplast diversity as well as the phylogenetic relationships of eight species of saffron including four spring-flowering and five autumn-flowering species from different parts of Iran, using the nuclear barc...
متن کاملIdentification of Crocus sativus and its Adulterants from Chinese Markets by using DNA Barcoding Technique
Background: Saffron (Crocus sativus L.) is a common but very expensive herbal medicine. As an important traditional medicine, it has an outstanding effect in treating irregular and painful menstruation. Recently, the over-demand tendency of saffron results in an unusual phenomenon in the medicinal markets. Adulterants and saffron-like substitutes are intentionally mixed into medicinal markets a...

Random amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP) markers were used to assess the genetic diversity in 31 species of mangroves and mangrove associates. Four AFLP primer combinations resulted in the amplification of 840 bands with an average of 210 bands per primer combination and 11 RAPD primers yielded 319 bands with an average of 29 bands per primer. Th...
